What is horse serum used for?
Gibco horse serum is donor-herd collected in New Zealand and can be used as an alternative to fetal bovine serum.Horse serum has been cited for use in several specific cell culture applications:Support of in vitro antibody response to SRBC in place of FBS [1]Production of C2 heterokaryons [2]Isolation and culture of primary neural cells [3]Promotion of myogenic differentiation [4]Supplement in mycoplasma broths and agars [5]In addition to horse serum as a lower cost alternative to FBS, Thermo Fisher Scientific also offers normal horse serum that can be used as a blocker solution in immunoassays.Gibco horse serum is available in both heat-inactivated and non-heat-inactivated formats.
Horse serum vs FBS
Horse serum can be used as a viable FBS alternative in some cell culture applications. One key horse serum benefit compared to FBS is its lower cost. However, horse serum composition does differ from FBS composition, with horse serum tending to exhibit lower growth factor and higher immunoglobulin levels than FBS [6]. Therefore, due to differences in FBS and horse serum, your specific experiment and needs should be carefully considered when deciding whether to use horse serum.
Horse serum collection, manufacturing, and testing
Collected from donor herds in New Zealand, each lot of horse serum is tested for its ability to support the growth of Sp2/O-Ag14 (murine myeloma) cells in control medium containing the test lot of serum. Results are compared to those obtained using control growth medium with a previously characterized reference serum. In addition, Gibco horse serum is EIA (equine infectious anemia) tested.

普諾賽特級胎牛血清具有非常卓越的促細胞生長能力,適合培養(yǎng)腫瘤細胞,原代細胞,部分干細胞和嬌貴細胞。
血清如何保存, 可以放多久?
貯藏溫度≤-15℃,可長時間保存; 2~8℃存放時,請勿超過一個月。
血清溶解以后,出現(xiàn)沉淀怎么辦?
若您欲去除這些絮狀沉淀物,可以將血清分裝至無菌離心管內,以400-600g離心5min,上清液即可加入培養(yǎng)基內一起培養(yǎng)。 我們不建議您以過濾的方法去除這些絮狀沉淀物, 一方面它可能會阻塞您的過濾膜;另一方面,過濾血清這種行為可能會導致血清中部分營養(yǎng)成分的流失。

細胞培養(yǎng)中“低調的高手”——馬血清 - 知乎切換模式寫文章登錄/注冊細胞培養(yǎng)中“低調的高手”——馬血清靈兒細胞培養(yǎng)中的馬血清說起細胞培養(yǎng)用的血清,大多數(shù)人都會想到胎牛血清、新生牛血清,對于馬血清卻知之甚少。今天就給大家介紹一下血清中“低調的高手”——馬血清。一、馬血清作用細胞培養(yǎng)級的馬血清,經(jīng)過多次過濾,去除了許多大分子蛋白,但保留了血清中促細胞生長的活性因子,表現(xiàn)出與胎牛血清和牛血清的類似的特性,因而能用于細胞培養(yǎng)。馬血清可以刺激成纖維細胞分化,以及細胞外基質 (ECM) 、細胞因子的合成。另外培養(yǎng)神經(jīng)細胞和心肌細胞、誘導生成樹突狀細胞等實驗均可用馬血清替代胎牛血清。因此,馬血清通常適用于那些研究有絲分裂后,細胞關于生長因子和激素等問題的實驗?;蛟S,我們可以這樣說,從現(xiàn)有研究來看,馬血清更適合用來研究特定種類細胞的分化。實際實驗應用方面:馬血清已被用作腦皮質神經(jīng)元-膠質細胞培養(yǎng)的一種必添加物;大鼠的 PC12 細胞,在培養(yǎng)過程中,也需要在基礎培養(yǎng)基中添加馬血清作為補充劑。1. 馬血清可用于細胞培養(yǎng)和干細胞的誘導分化,例如:(1)RPMI-1640培養(yǎng)液加馬血清培養(yǎng)臍帶血單個核細胞。(參考文獻:李春莉, 王建偉, 姜蓉, et al. 人參總皂苷誘導紅系血細胞增殖的信號轉導[J]. 中國組織工程研究, 2009(23):4568-4572.)(2)原代培養(yǎng)神經(jīng)細胞。有人用含10%馬血清的DMEM,分離培養(yǎng)BALB/c小鼠原代腦神經(jīng)細胞。發(fā)現(xiàn)和添加2%神經(jīng)細胞生長劑B27的Neurobasal培養(yǎng)基相比,含10%馬血清的DMEM培養(yǎng)基更節(jié)省成本,方便廉價。(參考文獻:李丹, 呂耀中, 王微,等. 兩種培養(yǎng)基分離培養(yǎng)BALB/c小鼠原代神經(jīng)細胞方法的比較[C]// 中國北方實驗動物科技年會. 遼寧省實驗動物學會, 2015.)(3)誘導脂肪干細胞向成肌細胞分化(文獻來源:劉杰,傅強.馬血清在脂肪干細胞成肌誘導中的作用[J].中國組織工程研究, 15(14))(4)培養(yǎng)骨髓基質細胞。與胎牛血清相比,馬血清對小鼠骨髓基質細胞的促增殖作用更好。(文獻來源:王改琴. 當歸多糖對小鼠骨髓基質細胞黏附分子表達及其黏附功能的影響[D]. 學位論文.重慶醫(yī)科大學, 2008.)(5)添加20%馬血清的L-DMEM培養(yǎng)液可誘導人骨髓間充質干細胞分化為脂肪細胞。(文獻來源:趙文靜,陳亞潔,劉薇.人骨髓間充質干細胞分離培養(yǎng)及生物學特性研究.中國現(xiàn)代醫(yī)學雜志.2006.16(18):2744-2747)2. 馬血清添加到微生物培養(yǎng)基中,可用于培養(yǎng)微生物,如:(1)馬血清添加到培養(yǎng)基中用于支原體的培養(yǎng)。在《獸藥典》中,含馬血清的支原體培養(yǎng)基被用于獸用疫苗的支原體檢查。用KM2培養(yǎng)基(含馬血清)還可培養(yǎng)豬肺炎支原體。(文獻來源:胡茂志 焦新安,等.豬肺炎霉形體單克隆抗體的制備與鑒定.《中國獸醫(yī)科技》 2004年12期)(2)馬血清加入到固體和液體培養(yǎng)基中,可用于培養(yǎng)幽門螺桿菌。并且,和羊血清和牛血清比較,添加馬血清的培養(yǎng)基更易于分裝和保存。(文獻來源:劉琳娜, 丁士剛, 張靜,等. 幾種培養(yǎng)幽門螺桿菌方法的比較[J]. 胃腸病學, 2012, 017(004):240-241.)3. 馬血清及激素可共同誘導建立家兔股骨頭缺血壞死的模型。(文獻來源:洪嵩, 張?zhí)旌? 非創(chuàng)傷性股骨頭壞死模型的血液流變學及脂質代謝[C]// 2009第一屆貴州骨科論壇.)二、注意事項與胎牛血清一樣,馬血清在使用過程中也應注意幾個原則:1、避免反復凍融;2、三步法解凍,及時分裝;3、配制好的完全培養(yǎng)基在4℃儲存不要超過7天。還有一個大家比較關心的問題,馬血清是否需要滅活?我們都知道,胎牛血清由于特殊的采血時間(5-8月胎齡),牛胚胎體內還沒有形成完整的補體系統(tǒng),因而不需要采用熱滅活的手段對補體進行滅活。而馬血清,在沒有特殊說明的情況下,一般采自成年馬匹。作為脊椎動物,成年馬也擁有完整的補體系統(tǒng)。因而,如果所進行的實驗中,涉及補體相關內容,建議大家在使用前對馬血清進行熱滅活,也可以直接購買商品化的滅活馬血清。一些比較成功的實驗案例中,所用到的馬血清,也是經(jīng)過熱滅活的。相關實驗請參考:https://www.ncbi.nlm.nih.gov/pmc/articles/PMC5111218/Ausbian馬血清為細胞培養(yǎng)級。它來自健康馬血液,為封閉系統(tǒng)內無菌采血過濾而成,為淡黃色澄清液體,超低內毒素,無溶血,無異物,無細菌、真菌、支原體、噬菌體等。產(chǎn)品描述1、每個批次均具有完整的檢測報告,其中含有下列檢測:1)理化檢查包括外觀、pH值、滲透壓、血紅蛋白、總蛋白、內毒素、蛋白顆粒度等2)無菌檢查含細菌真菌檢查、支原體檢查、噬菌體檢查,確保不含這些外源因子。3)病毒檢查牛病毒性腹瀉病毒(BVDV)為陰性。2、極低內毒素。3、提供試用,養(yǎng)細胞更安心。發(fā)布于 2022-05-15 21:58細胞死亡細胞細胞學?贊同 3??添加評論?分享?喜歡?收藏?申請

淺談「馬血清」在細胞生物學中的作用 - 知乎切換模式寫文章登錄/注冊淺談「馬血清」在細胞生物學中的作用愛生物的小蟲子深耕在生物圈內的科研狗細胞培養(yǎng)用Ausbian馬血清馬血清經(jīng)過多次過濾,祛除了許多大分子蛋白,但保留了血清中促細胞生長的活性因子,因而能用于細胞培養(yǎng)。相關科學研究1、馬血清可用于細胞培養(yǎng)或誘導樹突狀細胞,且細胞生長、增殖狀況良好。有研究人員分別用胎牛血清和馬血清培養(yǎng)馬單核細胞來產(chǎn)生樹突狀細胞(eqMoDC),結果發(fā)現(xiàn),馬血清馬血清生成的eqMoDC與FBS生成的eqMoDC沒有區(qū)別。然而,與馬血清補充培養(yǎng)基一起孵育的eqMoDC在從單核細胞分化過程中表現(xiàn)出更具特征性的樹突狀細胞形態(tài)。在用馬血清培養(yǎng)的eqMoDC中也觀察到細胞活力的顯著增加。此外,發(fā)現(xiàn)在馬血清存在下產(chǎn)生的eqMoDC在功能性T淋巴細胞啟動能力方面優(yōu)越,并且引發(fā)的非特異性增殖顯著減少。參考文獻:李春莉, 王建偉, 姜蓉, et al. 人參總皂苷誘導紅系血細胞增殖的信號轉導[J]. 中國組織工程研究, 2009(23):4568-4572.Ziegler A, Everett H, Hamza E, Garbani M, Gerber V, Marti E, Steinbach F.
